Spiral Timeline

        The spiral timeline represents the iterative and upward trend of iHP's development over time.Our 'Spiral Timellne"Is Insplred by two classlc concepts:the mathematlcal princlple of the "Flbonaccl sequence"that gives rise to the Golden Spiral,and the philosophical concept of "spiral ascent."The Fibonacci sequence re- flects the rhythmlc and systematic nature of IHP's closed-loop process,ultimately resulting In a splral upward movement on the axis.The philosophical concept of spiral ascent"embodies the convoluted de- velopment process of IHP.We have explored varlous paths,many of which have falled for varlous reasons. However.overall,we have continuously iterated and upgraded the content of experiments,modeling,and HP through many Interactlons In IHP,ultimately presenting a spiral upward motlon.In terms of signlfl- cance,our spiral timeline reflects our continuous pursuit of improvement in terms of requirements,sci- ence,Implementation,business,ethics,and Innovation Iteration.Additionally,It explains the entire opera- tion process of AFMU-China,and you will embark on a journey of human practice within this spiral time- line.If you only want to highlight the milestones and brainstorming of our joumey of human practice,we recommend reading the information in each small window and the milestones/brainstorming.

SRIP (Swiss Roll of integrated HP)

        SRIP,short for "Swiss Roll of integrated HP,"is inspired by the snack called "Swiss Roll."The Swiss Roll is characterized by layers wrapped around each other.similar to the layered and enveloping nature of our iHP closed-loop system.Thus,we named it SRIP.We have visualized almost all aspects of iHP's work and integrated them into this structure,which narrates how interactions with stakeholders,experts.profes- sors,and commercial companies shape our project plans and solution designs,helping us iterate our proj- ects.It connects all our human practice offorts to ensure that SRIP will have a significant and responsible impact on all domains.Additionally.it explains how SRlP should be implemented in the real world.
        The SRlP feedback loop consists of three interconnected closed loops:a core pentagram,a middle circle. and an outer rectangle.representing the lterative rigor of each layer of thought In the IHP design process. Entrepreneurship
        We've brainstormed and determined the direction for commercializing our product.We contacted com- panles such as GEGE Wu Blotechnology Co..Ltd.and Senmlao Blotechnology Co..Ltd.Through in-depth communication with them and leamning from them.we learned about the work we needed for product commercialization,the shortcomings in the design of the initial commercial plan,and how to make it more promotable.Based on these insights.we made the following iterations:

HP:

1.

Created a commercial plan that included more policy advantages and industry [Business plan]

2.

Designed a product user manual [Product instruction manual] to ensure accurate and error-free implementation of the reagent kit in every detail.

3.

Considering the users,including the elderly and visually impaired,we designed a "senior version prod- uct user manual" [Elder version of the product manual] based on the2O22"Design and Service Reference Guide for Aging",which reflects iteration in aspects such as font size.font design,and manual color

Experiment:

1.

To reduce the cost of the reagent kit,we used a lower-cost T4 DNA ligase to cyclize the probe.We also explored the optimal concentration of Cas12a to minimize the enzyme concentration without affect- ing reaction activity and reducing costs.

2.

Designed a honeycomb-like DSN vector for pursuing reuse of DSN in the future.

3.

To maximize the application scenarios of our reagent kit,all enzymes used in the kit can utilize its func- tions at room temperature or body temperature.

Modeling:

In the hardware part.we changed the initial design of having A and B solutions react separately in two tubes and finally decided to have them mixed in one tube with a semi-permeable membrane,which reduced consumable costs.

Activity Outcome:  Through this brainstorming session, the members gained a deeper understanding of each other. They actively brainstormed and gained a better understanding of synthetic biology and iGEM. Based on their interests, they proposed preliminary ideas related to testing, treatment, and other aspects. During the process of listening to others' ideas and presenting their own ideas, the members received valuable opinions and suggestions.

They presented a brief report on their preliminary project and continued to discuss and exchange ideas on the issues raised in the report. Ultimately, the decision was made to participate in the competition with the project on metabolic syndrome testing.

Activity Purpose:   Through an expert interview, understand the social status, hazards, and prevention and treatment of obesity.
Activity Outcome:   Professor Yang Ruihua introduced the social status and hazards of obesity, as well as scientific methods for preventing and treating obesity. He emphasized three key approaches to scientific weight loss: eating well, exercising reasonably, and reducing stress. The interview served as an effective means of dissemination. Additionally, Professor Yang provided valuable suggestions for our project's initial concept. For example, he recommended focusing on lipid metabolism-related molecules such as DID2, APOA, APOE, and certain genes, as they have strong implications in obesity and chronic diseases. This suggestion greatly inspired our project design.

Activity Purpose:   Understand the public's awareness of obesity and metabolic syndrome, as well as the real needs of society.
Activity Outcome:  Through more than 300 feedback questionnaires received online, the statistical results showed that about 45% of people considered themselves overweight or obese, and over 60% of people believed that obesity can lead to low self-esteem and other emotions. Approximately 98% of people believed that obesity can harm their physical health and are willing to lose weight. This is also the background reason for our firm choice of this research topic.

Activity Outcome:  We randomly interviewed 8 passers-by, including people of different genders, ages, heights, and weights. Most of them had experiences of being overweight or obese and expressed anxiety about obesity. More importantly, they provided valuable suggestions for the product we are going to design: they hope the price of the testing kit is not too expensive; they hope the sensitivity of the test can be relatively high; they hope the testing process is not too complicated.

Activity Purpose:   In-depth investigation of metabolic issues related to obesity from a clinical perspective.
Activity Outcome:  Professor Ji Gang explained the diagnosis and outpatient treatment methods for metabolic diseases caused by obesity. He mainly introduced gastric greater curvature resection surgery for patients with a BMI > 32 who did not respond to exercise or medical treatment. In addition, Professor Ji introduced the brain-gut interaction and discussed the potential mechanisms of obesity. Finally, Professor Ji recognized the prospects of our product and expressed willingness to introduce it to patients in the future.

Activity Purpose:  To discuss and determine the amplification and conversion methods of a signal in a multi-level amplification project.
Activity Outcome:   The students proposed various ways of presenting the final signal, such as colloidal gold colorimetry, hydrogen peroxide bubble display, and test paper display. They actively compared different signal presentation methods and exchanged ideas with relevant experts. Finally, colloidal gold colorimetry was chosen as the final signal presentation method for our project.

Activity Outcome:   During the experimental process, significant errors were observed in the results of the wet and dry experiments. The team conducted a brainstorming session to analyze the details of the process and consulted experts from Tsinghua University. As a result, improvements were made to the dry experiment process, and a reasonable model was established to assess system efficiency.
