Background
        Our main hardware is a home fast test kit for representative RNA of metabolic syndrome Fast Amplified Tace-RNA test kit,FAT test kit.The kit is mainly composed of three functional modules,namely sample collection module.reaction module A and reaction module B.two of which are separated by a thin film.
Composition
        The sample collection module,which containing the universal buffer,collects blood from the subject's fin- gertip through the blood collection needle included in the kit.
        The reaction module A contains two biological elements-the DSN enzyme and the nucleic acid probe. DSN enzymes can degrade the DNA strand in a DNA-RNA hybrid double strand,but has no effect on sin- gle-stranded nucleic acid molecules.We fixed DSN enzymes on the side wall of reaction module A by im- mobilizing enzyme technology.Nucleic acid probe can recognize specific MiRNA,and is a DNA-RNA hybrid loop single-strand probe.
        The reaction module B contain Cas12a protein to be activated,assistant DNA,and inhibit DNA-sgRNA hybrid double strands,which are anchored to the sidewall.The role of assistant DNA is to assist the activa- tion of Cas protein.Two small prominent single strands in the inhibit DNA are Cas12a's cleavage sites.

Principle
        After the sample collection module collects the blood and diluted it with universal buffer.the solution enters the reaction module A.MiRNAs in the blood would bind to the complementary region in the probe and form local DNA-RNA hybrid double strands through base complementary pairing.Then,the DSN en- zymes will degrade the DNA In the hybrld double strand and break the probe cicular structure.The MiRNAs in the blood and the unmatched RNAs in the probe are released.and the released MiRNAs carry out a new round of reaction to realize signal amplification and reuse.At the same time,the released un- paired RNA sequences self-fold to form mature sgRNAs,which will enter the reaction module B for the next reaction.
        When the solutlon enters reactlon module B.sgRNA will form a ternary complex with Cas12a and assls- tant DNA which can trans-cleave the prominent structure of the inhibit DNA and release the sgRNA se- quence.The released sgRNA will combine with Cas12a to be activated to carry out a new round of reac- tion and achieve exponential amplification of the signal.
Instruction
        Subjects can collect their fingertip blood,saliva or other body fluids containing RNA through the sample collection module,and transfer them to the reaction module A.When the body fluids react with the solu- tion in A for about 1 minute,subjects can use the matching needle to break the film between module A and module B,so that all the reaction llquld enters module B for the next reaction.The optimal detection time is about 9 minutes.Finally,the subject should take a photo of the reaction solution and upload it to our matching APP for result evaluation,which will complete the home test.The whole process takes about 20 minutes,which is very simple and fast and helps to monitor the health status of residents at any time.
