We obtained the wild strains with good ability to tolerate saline and base environment, they are Brevibacterium casei G20, Bacillus haynesii P19, Micrococcus luteus R17, Halomonas bluephagenesis TD01, Halomonas campaniensis LS21 and Enterobactercloacae RS3. We find their function genes for the tolerance. Then we constructed 21 recombinant plasmids. We have tested SURFB, and the surfB gene is very effective in improving the E. coli’s ability of function of model cells (E.coli). We also invited Prof.Li Chun's lab at Tsinghua University to test surfB and received good results. Here is their evaluation.

They said“The P19panC and LS21sufB genes used in the experiments showed significant enhancement in improving the alkali tolerance (pH=7.5-8.5) of E. coli BL21, with the enhancement effect of LS21sufB being more obvious.”

2 Gene mining system

To identify the common genetic elements differentially expressed in transcription, translation and metabolism that were commonly altered by the six salt-tolerant bacterial strains in response to salinity stress, we used p < 0.05 and |log2FC| > 0 as screening thresholds for the differentially expressed genes and differentially expressed proteins.

If a team has similar needs in the future, they can learn from these. This methodological process can be applied to any scenario where you want to mine genes, and we welcome anyone to talk to us or use this process!
