Materials
On this page, you will find information about the plasmids, strains, primers and DNA sequences we used. For the kits used for our experiments, the references are indicated on the protocol page.
Plasmids
- • pET21a (+): this plasmid was used as a backbone for almost all our constructs, as it allowed us to express all the proteins we needed. This expression is dependent on a T7 promoter. It confers ampicillin resistance. pET21a (+) was a gift from Ambre Jousselin. See the plasmid map on Snapgene here.
- • pET26b pelB-HER2-tev stops: this plasmid was used to express and produce the VHH anti-HER2. It contains a pelB leader sequence to direct the produced protein to the periplasm for further extraction and a Histag. This device confers the kanamycin resistance. This plasmid was a gift from Adilya Dagkesamanskaya.
- • p0579: this plasmid was used to clone and express the following fragment: Nterminal of the T7 RNA polymerase - linker - VH and VL of Pertuzumab antibody. It confers spectinomycin resistance. It originally contains Nterminal of the T7 RNA polymerase - linker - FRB to which we added an SP6 promoter with an RBS upstream and a T7 terminator. p0579 was a gift from Jérôme Bonnet.
- • p0539: this plasmid was used to clone and express the following fragment: VH and VL of Trastuzumab antibody - linker - Cterminal of the T7 RNA polymerase. It originally contains FKBP- linker - Nterminal of the T7 RNA polymerase to which we added an SP6 promoter with an RBS upstream and a T7 terminator. It confers chloramphenicol resistance. p0539 was a gift from Jérôme Bonnet.
Strains
- • E. coli Stellar: (F–, endA1, supE44, thi-1, recA1, relA1, gyrA96, phoA, Φ80d lacZΔ M15, Δ(lacZYA-argF) U169, Δ(mrr-hsdRMS-mcrBC), ΔmcrA, λ–) used for transformation during In-Fusion® cloning (Stellar Competent cells from Takara, ref: 638952).
- • E. coli BL21 (DE3): (E. coli str. B, F–, ompT, gal, dcm, lon, hsdSB(rB–mB–), λ(DE3 [lacI lacUV5-T7 gene 1 ind1 sam7 nin5]), [malB+]K-12(λS) used to express and produce DhdR and anti-Her2 nanobodies. The production of the T7 RNA polymerase can be induced by IPTG. This was a gift from Cédric Montanier (TBI, Toulouse).
