In our initial design, we had chosen the tetrameric streptavidin as the linker protein for Silinker. However, we later realized that using tetrameric streptavidin might lead to signal crosstalk or spatial hindrance for the functional proteins when aiming for Silinker's signal response capability. We discovered the engineered monomeric streptavidin, known as mSA, which can bind to a single biotin molecule in its monomeric form. During our exchange with DUT-China at The Conference of China iGEMer Community (CCiC), DUT-China's project on single-molecule detection reminded us of the valuable application potential of monomeric streptavidin as a protein linker. This reaffirmed our decision to use mSA as a replacement for tetrameric streptavidin.
