Our entire project is divided into two phases, the precious metal phase and the heavy metal phase. The following is the protocol we have prepared for our own experiments, which includes some precautions for conducting certain experiments (including the use of homologous fragment knocking out genes, Gibson assembly, Gel extraction, Transformations, Colony PCR and normal PCR, Quick ligation and plasmid miniprep). And we have made certain modifications to the protocol based on the conditions of our laboratory.

With the help of instructors and advisors, we do our best to improve our experimental protocol. We hope it could be a helpful reference for future IGEM teams.
