1 Laboratory Safety Training Course

Before entering the lab, the teacher gave a wet lab safety lecture to the members. Students should know how to use the equipment, how to deal with hazards, and how to protect themselves. For example, after the end of the experiment, make sure that the experimental equipment and electronic products are closed, and the Windows, doors are closed without ventilation. And to ensure that students into the laboratory must wear laboratory clothing and gloves, to avoid corrosive, harmful chemical reagent pollution to students, and close the reagent bottle to avoid upset, affecting the experiment

After the lecture, the tutor led the members on a rigorous tour of the laboratory to learn about the laboratory environment. She shows the location of safety equipment, for example, fire extinguishers, emergency eye wash and emergency exits. The instructor trained the students on how to use the safety equipment and told us to always ask the teacher for permission before using the equipment.

2 Safety rules and regulations of the laboratory

2.1 Daily laboratory management regulations

  1. The laboratory is an important place for experimental verification of samples, and unrelated personnel are not allowed to enter the laboratory without permission. Customer visitors who wish to enter the laboratory must be accompanied by relevant personnel from the institute.
  2. Laboratory personnel must strictly implement the institute's management systems, strictly prohibit the storage of personal or unrelated to the test, and do not allow non-work-related activities in the laboratory
  3. Keep the laboratory and products, instruments and equipment clean, dress according to regulations, and strictly prohibit smoking indoors
  4. Except for laboratory personnel, without permission, no other person shall operate, use or move all instruments and equipment and products of the laboratory
  5. Without permission, the tester shall not accept products outside the institute for testing or arrange others to carry out test activities in the laboratory
  6. The laboratory should be civilized and tidy, and the instruments and equipment and products should be placed neatly in an orderly manner

2.2 Regulations on the management of the use of laboratory test equipment

  1. The purchase of laboratory instruments and equipment and their consumables shall be handled in accordance with the relevant procedures of the company, and the instruments and equipment purchased shall be inspected and handled for warehousing and use
  2. All instruments and equipment in the laboratory shall be managed, operated, maintained, maintained and operated by the tester
  3. Laboratory instruments and equipment shall not be lent to personnel outside the laboratory without approval, and if they are approved to be lent to personnel outside the laboratory, relevant procedures shall be completed in accordance with the provisions before implementation
  4. For instruments and equipment that lose repair and use value due to backward technology, serious damage, and high maintenance costs, the operator shall submit a written application for scrapping and report it to the relevant leaders of the company for approval
  5. Operate the equipment in strict accordance with the equipment operating procedures, and non-test personnel are strictly prohibited from operating laboratory equipment
  6. The maintenance of the equipment shall be carried out in accordance with the company's "Equipment Management Control Procedures"

2.3 Laboratory safety management system

  1. First, conscientiously implement the policy of "safety first, prevention first", and do a good job in laboratory fire and explosion prevention, poison prevention, moisture prevention, shower prevention and other work.
  2. Laboratory personnel shall be responsible for the safety situation in the laboratory and deal with problems in a timely manner
  3. The laboratory should be equipped with fire safety measures (natural firearms, fire sand, etc.).
  4. Before the experiment, the tester must be familiar with the performance of various instruments and equipment, conduct safety inspections, strictly implement operating procedures, and make necessary safety protection
  5. The power cord of the instruments and equipment in the laboratory must be checked frequently for damage, and when the circuit electrical equipment fails, the power must be cut off before the inspection can be carried out; Do not touch and unplug electrical outlets with wet hands
  6. After the test work, the power supply and instrument switch must be turned off. Before leaving work, laboratory managers must check the operating instruments and the doors, windows, and unused water and electrical switches of the entire laboratory, and make sure that they are closed
  7. Ensure that the technical requirements, operation specifications, test items and test data of the laboratory are complete, and shall not be disclosed or leaked to the outside without the consent of the general manager of the company.
  8. When laboratory operations are required on holidays or other rest times, the consent of the company's leaders is required and two people are arranged to participate

2.4 Laboratory Code

  1. The laboratory is an important place for scientific research, testing and teaching, and it needs to be kept quiet, tidy and orderly, and unrelated personnel are prohibited from entering without approval
  2. Staff should implement the post responsibility system, consciously maintain the work order of the laboratory, and strictly abide by the rules and regulations of the laboratory

  5. Flammable, explosive, corrosive, toxic and other dangerous articles shall be used and kept in strict accordance with regulations, and shall not be placed arbitrarily

2.5 Laboratory maintenance system

  6. Before the experiment, conduct a comprehensive technical performance inspection of the participating equipment, restore and adjust various parameters, carry out necessary maintenance, and carefully check the random accessories to make the equipment stable and reliable and in good condition

  8. Instruments and measuring tools must be verified periodically in accordance with regulations, and shall not be used beyond the verification period or unqualified

3 Specific lab work

In the disinfection of ultra-clean benches and the recovery of electrophoretic gel, ultraviolet (UV) rays were used. According to the World Health Organization (WHO), increased UV exposure can cause skin cancers, cataracts and immune system damage. Therefore, in our experiment, during the long-term UV operation, we made sure to wear protective clothing to block the penetration of UV light. Also, special glass was used for isolation during laboratory disinfection and sterilization

Suspensions and immiscible liquids could be separated using centrifuges. The centrifuge tubes needed to be placed symmetrically in the rotor body with even numbers. Check whether the upper cover of the centrifuge was locked tightly.

When using a scalpel to cut the gel, we made sure to wear rubber gloves to reduce the chance of hand injury due to the sharp tip

Microwave oven was used to prevent glass bottle overheating and bursting. Heat-resistant gloves were put on to avoid burns when touching hot glass bottles heated by microwave oven and centrifuge tubes heated by Thermo scientific digital shaking dry bath

When using an autoclave, we made sure that it remained closed until the steam was exhausted. When the pointer of the pressure gauge of the pressure cooker was above 0.05Mpa, the steam should not be released too quickly to prevent the pressure from dropping rapidly, the liquid would boil and overflow from the culture container.

4 Specific chemicals

6×DNA Loading Buffer is used in our experiment to complete the processing of DNA samples before polyacrylamide agarose gel electrophoresis. It consists of 30 mM EDTA、36%(v/v) Glycerol、0.05%(w/v) Xylene Cyanol FF、0.05%(w/v) Bromophenol Blue. Among them, Xylene Cyanol FF may lead to eye and respiratory irritation in some persons. What is more, Bromophenol could be harmful to people by inhalation, swallowing or skin absorption as well. Therefore, it is necessary to wear goggles, disposable gloves and White coat when operating this experiment.

As considered that ethidium bromide can be absorbed through skin and it is an irritant to the skin, eyes, mouth and upper respiratory tract, which can cause damages to our body to some extent, we decided to use YeaRed Nucleic Acid Gel Stain (10,000×in Water) instead, since YeaRed is a unique oily macromolecule that cannot penetrate the cell membrane and enter the cell. Not easy to evaporate and sublimate, and the human body will not inhale. Ames test results also show that YeaRed is completely non mutagenic at gel dyeing concentration, and is a safe and non-toxic nucleic acid dye (Yeasen Biotechnology (Shanghai) Co.,Ltd.).

When we carry out the process of DNA or RNA extraction and purification, it is hard to avoid using chloroform which may make people feel dizzy, fatigue and headache after short time inhalation (From Wikipedia). And so, it is encouraged to wear a mask while extracting or purifying DNA and RNA.

When sterilizing the explants, we were asked to use Sodium hypochlorite to clean the explants for ten minutes. However, high concentration of Sodium hypochlorite may cause skin ulceration and it has certain irritation to respiratory tract as well, so we need to be extra careful when using it.

Lastly, when preparing the culture medium, we need to use hydrochloric acid and sodium hydroxide to adjust the pH value to the desired range. But as is well known, both of these two chemicals have strong corrosiveness, so it is necessary to wear all protective tools and handle them carefully during operation.
