Basic laboratory rules

· Wear protective measures (lab coats, rubber gloves, masks) in time

· Do not eat, smoke or drink in the lab

· Be sure to pay attention to the precautions and signs on each piece of equipment

· Closed-toe shoes and shorts cannot be worn in the lab

· Remember the emergency security phone number

· You can't experiment without any other people

Experiment

Clean bench

· The tools used need to be disinfected with more than 75% alcohol

· Turn on the ventilator before use

· Turn on the light when in use

Cellular room

· Be sure to wear masks, gloves, protective helmets, and protective clothing

Materials

· The bacterial strains we used in this project is Escherichia coli DH5-alpha and BL21.

· HEK293 cell lines were used for function tests and all relevant experiments were conducted in a BLS-2 lab with appropriate practices and containment facilities.

· Plasmids α2A-AR-CT and Venus-K-Ras are gifts from Dr. Xu, pGEX-4T-1 is provided by SubCat and other DNA materials were orderd from the company Genscript.

Morality

· Don't assume anything

· Take note of what others have given you

· Do not use reagents that do not belong to oneself without permission from others

· Don't litter

· Return the equipment after completing the experiment

GLP

· Place excess solute on an additional weighing paper and do not put it back into the original solute bottle

· Clean the spoon before changing solutes

· The balance needs to be peeled (zeroed) before each use with a weighing paper

· The lid of the solute bottle should be reversed on the desktop

· After using the Pipette, setting it into its maximum capacity

· To avoid pollution, hands should not touch the inside of the container walls

· hands should not be held on the outside of the container walls.

· When transferring liquid, check the range of the Single Channel Pipettors

· After using the solvent, put it back into the ice

· Press the piston to the bottom when adding liquid

· When adding solvent for the second time, place the needle below the liquid level

· Spray disinfectant alcohol before benchtop

· Unsterilized parts should not be inserted into a sterile environment (especially long-term items such as pipettes)

· When the liquid pipettor is used up, adjust the maximum range (according to the remarks on the gun body)

· You can add more (there are other substances in the solvent)

· Before using benchtop, you need to open the ventilation system

· Label all reagents
