Designing New Composite Part Constructs.

For our contributions, we created the following composite parts:

Part ID Part Name Part Description
BBa_K4999000 pBAD driven T4 lysis cassette This part involves the T4 lysis gene that is induced by arabinose. This allows for controlled lysis of E. coli cells, which is useful in our application of cell-free ATPS systems.
BBa_K4999001 LuxR driven by constitutive promoter This part involves combining LuxR under a constitutive promoter, which creates the protein needed for quorum sensing. This part can be integrated with other Lux components to create a quorum sensing circuit.

Candidate For Best Composite Part

Our significant contribution was designing a composite part (BBa_K4999000) that involved an arabinose inducible lysis system. Our composite part consists of the following basic parts:

DH5alpha E. coli Bacteria were transformed with the above plasmid. Lysis was shown for Clone #2 in the figure below.
