In (BBa_K4630001-BBa_K4630120), we designed a series of basic and composite parts to fulfill the recording event, using the concept of self-targeting editing and self-recombination.

• To construct composite parts, we added three stgRNA sequences generated by our drylab as basic parts (BBa_K4630001-BBa_K4630003), and some components we need (BBa_K4630004, BBa_K4630005). Then, two sets of designs were created: one with each cassette assigned a unique barcode to ensure accurate recombination (BBa_K4630100-BBa_K4630103), and another without the need for barcode accumulation (BBa_K4630110-BBa_K4630115), resulting in a more compacted and extendable device. Moreover, we employed EGFP as the reporter of successful recording and developed a series of parts (BBa_K4630116-BBa_K4630120).

(BBa_K4630200-BBa_K4630202) involve the three plasmids we used to express Cas9 and Lambda-Red. They are in line with the progress of optimization.
(BBa_K4630300-BBa_K4630301) are the two optimized new parts of the EL222 system developed by directed evolution.
(BBa_K4630400- BBa_K4630403) are the parts we designed to perform sgRNA screening. As these parts are directly synthesized or Gibson cloned, they are assembly incompatible up to now.
