Cas9 and Lambda-Red
For Cas9 (BBa_K1218011) and Lambda-Red (BBa_K1433005), combining them, we conducted characterizations involving the editing efficiency of the genome and plasmids. What's more, we shared some technical insights learned from our experiments, which may serve as references for others.
EL222 system and pEL222
For the EL222 system (BBa_K2447501), we identified a missing fragment in the part's sequence. We registered a new part (BBa_K4630301) to rectify this error and conducted our own characterization. Furthermore, we performed directed evolution on pEL222 (EL binding site, BBa_M45400) in three directions: reducing leakage and raising/lowering, with details available in the new part we registered (BBa_K4630300).
Thiosulfate sensing system, Pluxr, pPcau
For the parts in “Applications”, as we didn't receive the synthesized sequences before the WiKi freeze day, we supplemented these parts with new information gathered from literature sources. We added the characterization of literature to the thiosulfate sensing system (BBa_K3733032), Pluxr (BBa_R0062), and pPcau (BBa_K2825002).
