-IN THE LAB
l When experimenters do some experiments or touch equipments of experiments in the lab, they should wear disposable lab gloves, lab coats and disposable mask.
l Lab coats and gloves MUST NOT be worn out of the laboratory.
l When using every equipments, experimenters should know operational procedures and considerations.
l Don’t operate lab equipments without permission.
l Don’t eat or drink in the lab.
l Don’t taste any solution and substances in the lab.
-AFTER EXPERIMENTS
l Wash hands when you finish experiments.
l Clean up equipments and lab bench.
l All disposable equipment or used bacteria will be collected centrally.
- OPERATION OF THE PIPETTE
l Work on the lab bench.
l Don’t touch the pipette tip by hand.
l When there are any liquid in the pipette tip, hold the pipette vertically.
l When using the pipette to aspirate the liquid, it should not be sucked quickly, which will cause the solution to be sucked into the pipette.
l In order to reduce the splashing of liquid, the liquid is transferred into the container, and the tip is entered into the solution (but according to the experimental requirements)
l The tip must be replaced each time the liquid is removed.
l After using the pipette, adjust the pipette to the maximum range.
- OPERATION OF THE CENTRIFUGE
l The centrifuge should be placed and used horizontally.
l Before using the centrifuge, place the sample in equilibrium in the centrifuge, and if it is only a single tube, the other tube should take an equal mass of water instead.
l The lid of the centrifuge should be closed tightly.
- OPERATION OF THE PCR MACHINE
l When using the PCR machine, the environment should be constant.
l The power supply used by the thermal cycler should be stable.
l There should be no cross-contamination between samples.
- OPERATION OF THE SHAKE MACHINE
l We shake the bacteria on a shaker because the container is airtight, which ensures that the fungi are not infected with each other.
l We make sure the temperature is constant.
Our team used two competent states of E. coli:BL21 & DH5a
l We do each step of transferring microorganisms or genes, lighting alcohol lamps and alcohol disinfection of instruments, all in a sterile environment.
l These bacteria will be a little harmful to our health, so we will disinfect them before use.
