I.Basic laboratory safety

 

* Wear lab coats before entering the laboratory

* Use a fume cabinet for our investigations and wear face masks involving dangerous or unpleasant gases.

II.Safety of experimental operation

 

* Proper use of the pipettor

(1) Set the knob to the scale position you want. Before returning it to the scale, you can first slightly exceed it. To prevent human mistakes, it should be noted that scaling over the limit is not permitted.

 

(2) When inserting the tip and performing aseptic procedures, be mindful of swift and accurate movements, avoid touching the tip with your hand, maintain the movement, and forbid wasteful motions. When operating normally, tighten up with the thumb and index finger, but avoid touching the tip with the hand.

 

(3) Apply finger pressure to the first area until the reagent is touched. For instance, to prevent the inaccuracy brought on by the influence of hydraulic pressure, insert the white tip only 1 mm below the liquid surface, the yellow tip 2-3 mm, and the blue tip 2-4 mm.

 

(4) Aspirate the reagent slowly into the gun tip, keeping liquid and excessive speed to a minimum.

 

(5) To make the liquid flow down the outer wall of the tip after removing the tip, first gently lean on the inner wall of the container.

 

(6) Gently push the tip to the initial piece of the target containers inner wall to squeeze out the liquid.

 

(7) Remove the tip from the target container using your finger, being careful not to spill the reagent. Slide the tip slightly up against the inner wall of the container to retain a tiny quantity of liquid at the tip.

 

 

*  Working safety in the clean bench

 

1. Properly place the workstation

It is crucial to set up the clean bench properly before usage. In order to assure stability, the workstation needs to be anchored vertically to the floor in an open area. To maintain the efficacy of the workbench, it is crucial to keep the surrounding area neat, clean, and free of dust and debris.

 

2. Keep it pristine

The working space must be kept neat and orderly when the ultra-clean bench is being used. Prior to usage, the bench must be cleaned, and it must remain clean at all times while in use. Disinfect hands with ethanol before putting hands in the clean bench.Clean it immediately away after studies are over to stop the growth of germs or other things. To maintain the efficiency and caliber of your job, keep your workspace neat and orderly.

 

3. Using the UV light properly

It is crucial to use the necessary safety gear when using the UV light incorporated into the clean bench since it may help destroy germs and viruses and emit radiation. It is vital to keep in mind that overusing the UV light can also result in UV damage, thus it is crucial to utilize the UV lamp properly.

 

4. Check handling procedures

Utilizing an ultra clean bench necessitates the use of handling skills, which are crucial. Experienced users can utilize the ultra clean bench appropriately without causing any harm. To guarantee that the proper method is followed, novice operators need to acquire the proper operating procedures.

 

5. Ensure appropriate ventilation

To make sure that the air in the space is conditioned, the clean bench has to be thoroughly ventilated. Therefore, when utilizing the clean bench, it is crucial that the ventilation system be in good functioning condition. To maintain the workstation in an atmosphere that is acceptable for usage, the hood must always be maintained open when in use.

 

6. Controlling how the device is used

You must pay attention to both the standardization of usage and the proper technique while utilizing an ultra-clean bench in addition to the correct method. To ensure the operation's safety, the operation must be carried out in line with the established processes, such as the usage of equipment specification process.

 

7. Prevent deterioration

the application of an ultra-clean table procedure to prevent overuse and waste. The ultra-clean bench will require energy due to its flexibility and the way it functions, and it is not at all practical to rely just on trash to gradually regulate the environment. A good job of saving must be done when consuming.

 

III.Ethics

There are no human or animal components or genes in our experiment. Additionally, all genetic alterations created through our procedure are only used for study.

 

 

IV.Safety of materials

The organisms we used for experiments were Escherichia coli only, including DH5α and BL21 and these were competent cells purchased from companies.

The temperature of the molten agarose gel may rise to 90 degrees Celsius. To remove the container from the heater, put on heat-resistant gloves.

The nuclear dye for the agarose gel might have safety risk causing cancer, so we are required to wear gloves and throw away the contaminants at once after finishing the lab into the specific bin.
