Throughout the project, we always experimented in the laboratory. We strictly carry out experiments in specific areas, such as chemical fume hoods, biological safety cabinets and so on. Besides, our laboratory is also equipped with emergency kits, eyewashes, etc., to further protect the safety of students. Finally, the laboratory also has strict rules and regulations. In the process of the project, we always abide by the rules and regulations of the laboratory.
Laboratory biosafety
All our operations are carried out on the ultra-clean bench, and the used medium needs to be sterilized and placed in the garbage can for unified treatment.
Laboratory non-biological safety
There are special collection and storage places for organic waste liquid and broken glass products, and professional personnel will deal with them in a unified manner.
Training
Before the experiment was carried out, our senior fellow apprentice trained everyone on laboratory safety.
Dress
During the experiment, we will wear our own experimental clothes. In addition, volatile organic solvents (such as methanol) were used in our experiments, so we will wear KN95 masks during the experiment to ensure our safety to the greatest extent. We also wore two layers of gloves during the experiment(the inner layer is a polyethylene glove and the outer layer is a nitrile glove).
Use of instruments
Before the formal test, the laboratory manager trained us for the use of the instrument. The team members already can use various instruments, and can skillfully and safely use instruments such as Autoclave.
The engineering bacteria we used in the experiment was E. coli. According to ATCC, E. coli belongs to the first category in biosafety, that is, it is almost not pathogenic to humans. In addition, we will unify the medium containing colonies after the experiment to strictly avoid the leakage of all strains after the experiment, thus affecting humans or other animals.
Toxins and parts
The parts and products used in the experiment are non-toxic or low-toxic products, and the combination of components will not produce substances harmful to human body or environment.
Transport of DNA elements
All kinds of DNA elements we used have been approved and supervised by the corresponding enterprises and government departments. There is no risk in the transportation process and they are completely safe.
Connection with HP
In the propaganda activities of HP to primary and secondary schools, laboratory safety was also preached, mainly involving the harm of microbial leakage and how to avoid microbial leakage after the experiment.
