We use P450 monooxygenase CYP107D1 (Olep) to hydroxylate deoxycholic acid (DCA) to produce the target
product 6β-OH DCA.

catalytic efficiency
Our experiment was designed to improve the catalytic efficiency of P450 enzymes through the following four strategies, respectively:
(1) Increasing the soluble expression of Olep for the hydroxylation of DCA in E. coli.
(2) Enhancing the heme supply in E. coli to increase conversion rate.
(3) Screening and modification of the optimal redox partners for Olep by sfGFP and semi-rational design.
(4) Construction of the optimal whole cell catalytic system for Olep to perform most efficient conversion.
