We have completed the Attribution Form and put it on our Wiki. This page describes what each team member contributed to the whole project as well as what external partner helped our project. It also describes our project timeline, indicating the time when different parts of the project work were initiated and completed.

Project Description

We have completed the Project Description and put in on our Wiki. In this part, we illustrate the goal of our project: degrading α-pinene in turpentine based on engineering bacteria. We believe that our project will be a useful application of synthetic biology, as it will address the pungent odors of oil painting pigment thinners in an environmentally friendly way. We described how the two systems, α-pinene oxidation system and α-pinene oxide degradation system, were designed and tested to achieve the goal of degrading α-pinene. Relevant literatures were cited to support our research.

We have completed the Contribution for our Wiki, which is important for future iGEM teams. This page summarizes the inspiration and innovation of our project and troubleshooting for experiments. We added new parts and composite parts to the iGEM registry to facilitate access and to be used by future teams. For experiments, we applied simplified aqueous-organic two-phase system, providing inspiration for future iGEM teams. We also described tips and troubleshooting for the purification of transmembrane proteins. In addition, comprehensive models of α-pinene degradation pathway and the co-culture of two strains of E. coli were created. We also summarized what we have done to achieve lab safety and how we designed to guarantee the product safety. Our workflow can be used as a reference for future iGEM team.

Engineering Success

On this page we wrote how we achieve engineering success via multiple engineering design cycles. A standard cycle follows the order of designing, building, testing and learning. For each part, including GLF protein purification, aqueous-organic two-phase system, and co-transformation, at lease two cycles were included. We finally successfully engineered our bacteria with the ability of α-pinene degradation.

We have done Human Practices to prove that our project is responsible and good for the real world. We made various of investigations from aspects of turpentine's odor, market condition, user group, business, and product form. We also communicated with stakeholders from different backgrounds, including practitioners, market, and academia, providing us with a new perspective on the challenges and possibilities of our project. Their feedback promoted us to reflect our existing results and integrate their needs into our project.

New Basic Part


We added a set of New Basic Parts on Registry of Standard Biological Parts. Here are our parts: BBa_K4940004, BBa_K4940006, BBa_K4940007, BBa_K4940008, BBa_K4940009, BBa_K4940013, BBa_K4940014, BBa_K4940015, BBa_K4940016. Some of these parts came from published work, while some were modified by us. These parts play different roles in the process of α-pinene degradation.

Integrated Human Practices

We set an individual page for Integrated Human Practices, where we actively considered how the world affects our work and our work will affect the world. Our project aims to solve the problem of the pungent odors of turpentine. To do this, we conducted activities with the loop between what we designed and what is desired. We communicated closely with many different stakeholders including practitioners, market, and academia. We also conducted online and offline surveys on pupils, students of related majors, users and other general public to investigate different perspectives. Their feedback provided useful guidance for our project. In addition, we worked on social media and set Science Column to disseminate science knowledge including biosafety. Hope our project can influence more social organizations and create a win-win potential in the future.

Safety and security

Our team is committed to establishing a safe environment for experiments and research. In addition to basic laboratory safety training, we pay attention to operational safety during experiments. We designed experimental ideas based on the principle of safety, including chemical safety and biosafety. On the design of product structure, we also considered monitoring techniques, control of the release and waste management. To better popularizing biosafety to the public while emphasizing the security of our products, we wrote a Biosafety Handbook. iZJU-China hopes that we can live in harmony with microorganisms and create a better home together.
