General safety rules

1. Always put on lab coat and rubber gloves before doing experiment

2. Remember to take off your coat and gloves when leaving the lab

3. Do not bring any reagent outside the lab

4. Do not bring any micro-organisms that may cause infection into the lab

5. Do not taste anything inside the lab without permission

6. Do not rub your eyes with gloves on

7. Do not eat, drink, or run inside the lab

8. Handle all glass equipment with care

9. Clean up the lab before leaving

Experiment safety rules

Clean Bench

1. Spray ethyl over everything that will be sent into clean bench

2. Open air fan inside the bench before use

3. Light the alcohol lamp inside while using

4. Open UV light for sterilization after use

5. Clean the rubbish can each time after use

Organism Safety

1. The strains we only used in our experiment are Escherichia coli DH5-α and BL21.

2. We didn’t use Staphylococcus aureus and Pseudomonas aeruginosa, we replaced the test strains with the E. coli within pUC57-femA and pUC57-GbcA, respectively.

3. Cas12a plasmid was provided by SubCat and other genetic materials, including sgRNA and primers were ordered from companies Genscript and Azenta.

Gel Electrophoresis

1. Do not touch the flask with heated gel with bare hands

2. Do not look straight into the ultraviolet light when cutting the gel

Centrifuge

1. Always remember to balance the weight of reagents before starting the centrifuge

2. Clean up all the water inside the machine before cooling it down to 4℃

3. Close the lid after adding the reagent before starting the machine

SDS-PAGE

1. Be aware that CCB dye solution is poisonous due to the acetic acid it contains

2. Recycle the used CCB dye inside a fuming cupboard

Pipette

1. Do not turn the equipment upside-down when there is liquid inside

2. Do not set a liquid extraction amount greater than its maximum volume

3. Do not dip the pipette into any liquid without a pipette tip on

Refrigerator

1. Do not touch the walls of -20℃ fridge with bare hands

2. Always lock the door of the fridge after use

Disposal safety rules

1. Dump waste liquid and waste consumables separately

2. Do not mix organic waste materials like bacteria solution with other wastes

3. Clear lab rubbish can every day
