Here are all of the contributions that we made to iGEM's repository and group as a whole. It's been so great working with everyone from this growing community.
Documented and characterised a new dapA part for E. coli. Although there is an existing part for dapA this is from Bacillus Subtilis and has a sequence diversion of 50% from our part (BBa_I718005 ). dapA is involved in lysine biosynthesis and we confirmed with digestions and colony PCR correct assembly and devised a ninhydrin assay for quantifying the levels of amino acids in our cells as a rapid test before further metabolomics. We showed that this part when overexpressed does result in a significant increase in lysine. Future teams should seek metabolomic confirmation that the ninhydrin results correspond specifically to lysine increases and not other amino acids. Edinburgh sent samples for mass spec analysis but unfortunately, results did not arrive before the wiki freeze.
We made an educational leaflet for the general public about cyanobacteria, genetic engineering, the risks of GMO release and containment strategies using our projects as examples.
The idea for the Biologix was raised by the KCL team at the UK meetup organized by Edinburgh. The project was an educational collaboration between teams Kings College London, Warwick, Cambridge, Oxford, Edinburgh and Exeter. The aim was to design a PowerPoint lecture and textbook series aimed at English-speaking high school students. These materials would then be available for distribution to schools. Edinburgh has not focused on Education as an award, so we did not contact high schools for potential distribution.
KCIS reached out to us about doing a postcard collaboration where teams around design and send them a postcard depicting their projects. We did not end up sending the postcard, but the design was emailed to them online.
IISER Pune approached us about a collaboration to promote sustainable and healthy living, by suggesting a joint Instagram post where teams around the world send photos of them biking to school instead of taking private vehicles/public transport.
To celebrate World Nature Conservation Day, SVCE Chennai invited us to plant a tree on our campus and take photos for them. We coupled this into a fundraising event by inviting our professors to dress up as characters in The Lorax with us while we planted the tree.
We organized an online meeting to get in contact with other UK teams, exchange preliminary project ideas and discuss potential collaborations.
We started with each team introducing their projects, followed by a breakout room rotation where individual teams could ask each other questions and discuss potential collaborations. In this meeting, we discussed a potential software collaboration with the Cambridge dry lab team.
In celebration of World Biodiversity Day, team UniCrete invited us to an Instagram post collaboration showcasing the national animal of our country.
The work and results demonstrate that the newly developed CyanoGate system is user-friendly and can be a good option for any scientist interested in introducing new genes or operons into cyanobacteria. This will provide an example for future iGEM teams of molecular cloning using CyanoGate in a wide variety of genes.
This year, the 2023 iGEM Edinburgh Team has created a collection of parts centred around CyanoGate. We have submitted a few new basic parts (for example BBa_K4849001) and used these to create a range of composite parts. These submissions contribute to the small yet growing collection of parts that can be expressed in cyanobacteria. CyanoGate was the primary technique we used to assemble our DNA, and our experiences and designs will be invaluable to future teams hoping to work with cyanobacteria. We hope that our contribution makes cyanobacteria more accessible to work with, and that future teams will use our parts in their CyanoGate designs!
