Overview

As part of the ICARUS system, we decided to build a construct with the most optimal secretion. To achieve this, we decided to test different combinations of signal peptides, promoters and cargos. In this process, we aim to select a combination that will secrete soluble cancer antigens with the highest secretion efficiency possible. As such, we have constructed and utilized a variety of basic and composite parts in the build-up of our system.

Basic Parts

Our team selected multiple signal peptides for secretion of proteins in mammalian cells. Through literature search we identified most efficient signal peptides and decided to study them. In addition to these parts, we selected the HA tag (BBa_K1150016) and BM40 (BBa_K2170214), which both are reported in the registry.

Composite Parts

As our end goal, we want to create a library of functional expression units that are capable of secreting protein cargos of choices with desired secretion efficiency. There is a number of composite parts that we have not yet tested, so we refrained from including them in this list until we gather more reliable data on their use.
