ASTERISK-HBA1 / ASTERISK-HBB + T7 promoter and terminator
Figure 1: Circuit diagram of the ASTERISK-HBA1 (BBa_K4877021) and ASTERISK-HBB (BBa_K4877022) composite parts. The difference between these parts lies in the 5' and 3' UTRs. The ASTERISK-HBA1 contains UTRs from the human alpha globin 1 gene, and the ASTERISK-HBB part contains UTRs from the human beta globin gene.

BBa_K4877010 Coding sequence (peptide) P2A self cleaving peptide. This peptide isolates the mCherry fluorescent marker from the cloning site.
BBa_K4877016 Cloning site Sensor cloning site for inserting a sensor sequence. Sensors can be inserted by HindIII digestion followed by Gibson assembly.
BBa_K4877011 Coding sequence (peptide) T2A self cleaving peptide. This peptide isolates the cloning site from the payload sequence (mNeonGreen).
BBa_K4877017 Coding sequence (reporter) mNeonGreen fluorescent protein; surrogate payload reporter.
BBa_K4877012 Coding sequence (peptide) E2A self cleaving peptide. This peptide isolates the payload sequence from the MCP-ADAR2dd sequence.
BBa_K4877014 Coding sequence (enzyme) MCP-ADAR2dd(E488Q) coding sequence.
BBa_K4877007 3' UTR 3' UTR sequence of the human alpha globin 1 gene. This UTR is used in the ASTERISK-HBA1 part. The sequence does not contain the PolyA-signal.
BBa_K4877009 3' UTR 3' UTR sequence of the human beta gene. This UTR is used in the ASTERISK-HBB part. The sequence does not contain the PolyA-signal.
