Overview

New Basic Parts

We registered and uploaded basic parts BBa_K4606001, BBa_K4606002, BBa_K4606003, BBa_K4606004, BBa_K4606005 and BBa_K4606006.
We also uploaded basic parts BBa_K4606007 as a backbone and proved it worked as we expected.

New Composite Parts

We have designed three composite parts (BBa_K4606010, BBa_K4606011 and BBa_K4606012) in order to overexpress the corresponding genes in cells.

BBa_K4606002 Firefly-luciferase Coding Firefly-luciferase is a typical enzyme protein consisting of 550 amino acids and can catalyze the oxidation of luciferin to oxyluciferin and then emit bioluminescence.
BBa_K4606003 SMAD4(wild type) Coding SMAD4 is a transcription factor that has been reported to be closely associated with osteoarthritis. This 414bp-long fragment of SMAD4 is the head of our recombination device, which has the paired base sequence of miR-3074-5p according to the online repository.
BBa_K4606004 SMAD4(mutant type) Coding SMAD4(MUT) was generated by mutating the sequence part of SMAD4(WT) that is paired with miR-3074-5p.
BBa_K4606005 miR-3074-5p coding sequence Coding As the coding sequence of miRNA, it can be cleaved to miR-3074-5p after transcription.
BBa_K4606006 miR-3074-5p RNA miRNA3074 is over-released in the obese state and is associated with osteoarthritis.
BBa_K4606007 Vector-AmpR Plasmid_Backbone Vector-AmpR is a vector plasmid backbone. With this device, we can insert the coding sequence of a novel protein.
BBa_K4606010 Vector-ZsGreen-miR-3074-5p-AmpR Plasmid We loaded the genes encoding miR-3074-5p into plasmid vectors. In this way we were able to over-express the miRNA in the target cells.
BBa_K4606011 Vector- Luciferase-SMAD4(WT)-AmpR Plasmid We integrated the fragment sequence of the SMAD4(WT) gene in front of the luciferase gene and loaded them together into the plasmid vector. If the SMAD4(WT) sequence is successfully expressed, then the downstream luciferase gene will produce active luciferase.
BBa_K4606012 Vector- Luciferase-SMAD4(MUT)-AmpR Plasmid We integrated the fragment sequence of the SMAD4(MUT) gene in front of the luciferase gene and loaded them together into the plasmid vector. If the SMAD4(MUT) sequence is successfully expressed, then the downstream luciferase gene will produce active luciferase.
