
DNA sequencing
Following the RE pattern conformation (see result page), the clones of samples 1, 2, 5, and 6 were sent for DNA sequencing to determine whether the DNA sequences in the selected clones aligned with that of lumbrokinase gene. Overall, only clones in sample 6 had the correct DNA sequence.
In figure 1, the sequencing result of sample 6 offered valuable evidence regarding the success of the target protein production. To begin with, both the max score and total score reached a high value of 1338, indicating a strong alignment between the DNA sequences in our experimental clones and the lumbrokinase gene. Also, the E value for the sequences in our clones was recorded as 0, which is typically considered as a strong indicator of favorable homology matches. Lastly, it's worth noting that the result of percentage identity was 99%, indicating that most of the sequences within the selected clones were consistent with the lumbrokinase gene. Based on the evidence presented above, our team concluded that lumbrokinase was able to be successfully produced in our experiments so far.
